The In Vitro Effect of Adiponectin on Early Bovine Embryonic Development and Transcriptomic Markers of Oocyte Competence

چکیده

Objectives: The current study aimed to investigate the in vitro effects of physiologic concentration of adiponectin in follicular fluid of pre-ovulatory bovine ovarian follicles on: 1) the bovine oocyte maturation and the early developmental rates of bovine embryos in vitro; 2) the expression profiles of some developmentally important marker genes in the bovine cumulus cells and oocytes. Materials and Methods: The cumulus-oocyte complexes (COCs) were aspirated from abattoir ovaries and cultured in vitro in the presence/absence of the recombinant human (rh) adiponectin (5 μg/mL) and used for real time RT-QPCR and in vitro embryo production assays. Results: Adiponection had no significant effects on nuclear maturation and cumulus expansion rates (p > 0.05). The expression levels of hyaluronic synthase 2 gene (Has2) was significantly higher in the adiponection group compared to the control group (p = 0.014). However, the early embryonic development was not affected significantly in the presence of adiponectin. Adiponectin did not change the expression rates of adiponectin receptors or other markers of oocyte competence. Also, the expression rates of two important oocyte secreted factors (OSFs), bone morphogenetic protein 15 (Bmp15) and growth/differentiation factor 9 (Gdf9) were not affected (p > 0.05). Conclusions: Adiponection did not affect the bovine oocyte developmental competence and early embryo development in spite of a significant increase in the expression level of Has2 gene, a biomarker of the future oocyte developmental competence.

P-121: Assessment of Microtubule and Nuclear Status at Different Intervals of Bovine In Vitro Oocyte Maturation

Background: Mammalian oocyte undergoes a series of structural nuclear modulations during maturation in order to obtain full competence to support fertilization and early embryonic development. Microtubules are major cytoskeleton components and have pivotal modulators for chromosomal movement, and it has been shown that partial or immature spindle organization may compromise correct ploidy of th...

Effect of Different Harvesting Techniques on the Recovery and Quality of Bovine Cumulus Oocyte Complexes

This study investigated the effect of different harvesting techniques on the recovery rate and quality of bovine cumulus oocyte complexes (COCs) and their subsequent developmental competence including in vitro maturation, fertilization and blastocyst formation. The COCs were retrieved from slaughter ovaries using aspiration, slicing, or aspiration followed by slicing. The COCs were examined mic...

Background: Cryopreservation of farm animal embryos is nowadays a widely used routine technique. Success in cryopreservation of form animal depends on many factors including: freezing medium, types of cryoproctants, melting process, lipid content in ooplasm and oocyte quality It has been found that immature Oocytes with reduced reproductive performance or slaughtered at the end of their use are...

P-110: Effect of Increasing Amount of Oocyte Secreted Factors on Cumulus Expansion of Bovine Cumulus-Oocyte Complexes

Background: In vitro maturation is a good method to decrease cancer risk of superovulation by gonadotropin hormones. A paracrine effect of oocyte secretions on oocyte developmental competence is under investigation. Apart from oocyte maturation, ovulation in vivo requires a precise control of extracellular matrix modification. Cumulus cells secrete hyaluronan to form a muco-elastic extracellula...
